For most inground pools, as long as you prime your vac hose and set it up prior to attaching to the skimmer or to the vac plate, yes, the pool system (pump) should be on. If you have any air trapped in the line you will likely lose prime, so will need to give the pump a minute to reprime before you can begin to vacuum.
